Glycogen
synthase kinase-3α (GSK-3α) and GSK-3β are highly similar isoforms of serine/ threonine kinases that
regulate metabolic enzymes and transcription factors, which are responsible
for coordinating processes such as glycogen synthesis and cell adhesion.
GSK-3β activity is also required for nuclear activity
of Rel dimers, which mediate an anti-apoptotic response to TNFα in mice. GSK-3 catalytic kinase activity is controlled through
differential phosphorylation of serine/threonine residues, which have an
inhibitory effect, and tyrosine residues, which have an activating effect.
Growth factor stimulation of mammalian cells expressing GSK-3α and GSK-3β induces phosphorylation of Ser
21 and Ser 9, respectively, through a phosphatidylinositol 3-kinase (PI 3-K)-protein
kinase B (PKB)-dependent pathway, thereby enhancing proliferative signals.
Additionally, GSK-3 physically associates with cAMP-dependent protein kinase
A (PKA), which phosphorylates Ser 21 of GSK-3α or Ser
9 of GSK-3β and inactivates both forms. GSK-3α/β is positively regulated by
phosphorylation on Tyr 279 and Tyr 216, respectively. Activated GSK-3α/β participates in energy metabolism,
neuronal cell development, and body pattern formation. Tyrosine
dephosphorylation of GSK-3 is involved in its extracellular signal-dependent
inactivation.

保存條件:
Phospho-GSK-3α/β (Tyr216/279) Rabbit Monoclonal Antibody -20℃保存，Western一抗稀釋液-20℃或4℃保存，一年有效。Western一抗稀釋液優(yōu)先推薦4℃保存，長(zhǎng)期不使用可以考慮-20℃保存，但凍融可能會(huì)導(dǎo)致出現(xiàn)輕微的渾濁和少量不溶物。
注意事項(xiàng):
如果本抗體用于Western blot (WB)、**熒光(IF)、**細(xì)胞化學(xué)(ICC)等實(shí)驗(yàn)，請(qǐng)注意回收使用過(guò)的稀釋抗體。回收的抗體通常至少可以重復(fù)使用5-10次。稀釋后的抗體，包括已經(jīng)使用過(guò)的稀釋抗體，請(qǐng)4℃保存。
回收后重復(fù)使用的抗體，使用方法同新鮮稀釋的抗體。如果在重復(fù)使用過(guò)程中發(fā)現(xiàn)抗體出現(xiàn)輕微混濁現(xiàn)象，可以10,000g離心1-3分鐘，取上清用于后續(xù)檢測(cè)。如果回收的抗體出現(xiàn)明顯的絮狀物或長(zhǎng)霉長(zhǎng)菌等情況，則可以考慮廢棄該抗體。
提供的Western一抗稀釋液也可以用于**熒光(IF)、**組化(IHC)、**細(xì)胞化學(xué)(ICC)等適當(dāng)用途。如果希望獲得*佳的檢測(cè)效果，請(qǐng)考慮使用上述檢測(cè)專用的一抗稀釋液。
